【摘要】 目的分析中介体19亚基(Med19)表达与乳腺癌临床病理特征的关系。方法用RT-PCR检测Med19基因在6株人乳腺癌细胞ZR-75-30、MDA-MB-231、SK-BR-3、T-47D、HCC-1937及MCF-7中的表达。用免疫组化方法检测Med19蛋白在150例临床乳腺癌组织及癌旁正常组织中的表达并分析其与临床病理特征的关系。结果 Med19基因在人乳腺癌细胞株ZR-75-30、MDA-MB-231、SK-BR-3、T-47D、HCC-1937及MCF-7中均有较高丰度的阳性表达。乳腺癌组织中Med19蛋白阳性表达率为52.0%,明显高于癌旁正常组织的9.3%(P<0.01)。Med19蛋白的阳性表达与乳腺癌病理高分级相关(P<0.01),未发现与患者年龄、肿瘤大小、病理组织类型、TNM分期、雌激素受体(ER)、孕激素受体(PR)、p53有统计学意义的相关性(P>0.05)。结论 Med19在乳腺癌组织中高表达,可能在乳腺癌的发生发展中起重要的作用。更多还原

【Abstract】 Objective To investigate the expression of mediator complex subunit 19(Med19) in breast cancer and its relationship with clinicopathological features.Methods Real-time quantitative reverse transcription-PCR was used to detect the expression of Med19 gene in 6 human breast cancer cell lines of ZR-75-30,MDA-MB-231,SK-BR-3,T-47D,HCC-1937 and MCF-7.Immunohistochemical assay was performed to analyze the expression of Med19 protein in 150 matching pairs of breast cancer and tumor-adjacent non-tumor tissues.The association between Med19 expression and clinicopathological features of breast cancer was evaluated.Results Med19 gene was abundantly and positively expressed in all of 6 human breast cancer cell lines.The positive expression rate of Med19 protein in breast cancer was higher than that in tumor-adjacent normal tissues(52.0% vs.9.3%)(P<0.01).Med19 protein expression was significantly associated with the tumor of higher grade(P<0.01).No significant association was observed between Med19 protein expression and age,tumor size,TNM stage,estrogen receptor status,progesterone receptor status and p53 expression.Conclusion Med19 is highly expressed in breast cancer,which may play an important role in the development and progression of breast cancer.更多还原
