MK-801诱导出生早期大鼠精神分裂症模型的行为学研究
Study on the behavior of rat model of schizophrenia induced by MK-801 in early postnatal period
【摘要】 目的建立地卓西平马来酸盐(MK-801)慢性处理的出生早期大鼠精神分裂症(SZ)模型,探讨其神经行为学变化。方法 11只出生后6 d的SD仔鼠,腹腔注射0.3 mg/kg的MK-801(MK-801组),一天2次,连续注射2周;11只同龄的SD仔鼠腹腔注射等体积的生理盐水(NS组)。给药前称重1次,给药完毕后每隔2 d称重1次,直至实验结束。给药后1周,检测旷场和水迷宫等行为学的变化。结果与NS组比较,MK-801组大鼠体重显著降低(P<0.01),并至少持续至注射结束后4周。行为学检测结果显示,与NS组比较,MK-801组大鼠自发活动显著减少(P<0.01),其学习能力明显降低,学习速度减慢,空间记忆能力下降(P<0.05)。结论 MK-801诱导的出生早期SZ模型大鼠自发活动减少,能部分模拟SZ的阴性症状;其学习及记忆功能损害可以部分模拟SZ的认知功能损害症状。
【Abstract】 Objective To study the learning and memory characteristic of rat models of schizophrenia induced by chronic MK-801 treatment in the early postnatal period.Methods 22 SD rats 6 d after birth were divided into MK-801 group and normal saline group of 11 each,and the rats in the two groups accepted intraperitoneal administration of MK-801(0.3 mg/kg) or saline(0.3 ml/kg) twice a day for 2 weeks,respectively.Body weights of rats in two groups were measured before and every three days after injection,and continued up to the end of the experiment.After 1 week of injection endpoint,open field and water maze were detected in the rats of two groups.Results Compared with the normal group,body weights of rats in MK-801 group were gradually decreased during the injection process(P < 0.01) and at least continued for 4 weeks after the end of injection.The results of behavior showed that,compared with the normal saline group,the spontaneous activity in rats of MK-801 group significantly reduced(P < 0.01),and the ability of learn and memory significantly were declined in rats of MK-801 group(P < 0.05).Conclusion The decrease in the spontaneous activity of rat model of schizophrenia induced by MK-801 in the early postnatal period could partially imitate the negative symptoms of schizophrenia,and the dysfunction in learning and memory is similar to the cognitive impairment of schizophrenia.
