山楂叶悬钩子水提取物对转化生长因子-β刺激的肝星状细胞活化的抑制作用及其机制

【摘要】 [目的]探讨山楂叶悬钩子水提取物(RC-H)对转化生长因子(TGF-β)刺激的肝星状细胞(HSC)活化的影响及其机制.[方法]采用MTT法测定RC-H对大鼠肝星状细胞(tHSC/Cl-6)存活率的影响;采用TGF-β促活化tHSC/Cl-6细胞,蛋白印迹分析RC-H对TGF-β刺激HSC信号转导的影响.[结果]0~160 mg/L RC-H对tHSC/Cl-6存活率无显著影响(P>0.05),320,640,1 280 mg/L RC-H显著抑制tHSC/Cl-6的存活率(P<0.05).40,80,160 mg/L RC-H可抑制TGF-β活化tHSC/Cl-6细胞中α-SMA蛋白表达,下调TIMP-1蛋白表达,上调MMP-13蛋白表达,降低p-Erk和p-JNK蛋白表达,对pp38无显著影响.[结论]RC-H可通过介导MAPK信号通路中Erk和JNK磷酸化而抑制TGF-β刺激的肝星状细胞活化.

【Abstract】 OBJECTIVETo study the effects and its mechanism of aqueous extract of Rubus crataegifolius(RC-H)for activation of hepatic stellate cells tHSC/Cl-6stimulated by transforming growth factor-β(TGF-β).METHODSEffects of RC-H on cell viability of tHSC/Cl-6were detected by MTT,and on the signal transduction of tHSC/Cl-6activated by TGF-βwere detected by Western blot.RESULTS0—160 mg/L of RC-H had no obvious effects for the cell viability of tHSC/Cl-6(P>0.05),and 320,640and 1 280mg/L of RC-H had significant effects for the cell viability of tHSC/Cl-6(P<0.05).40,80and 160 mg/L of RC-H in tHSC/Cl-6activated by TGF-βinhibited the expression ofα-SMA and TIMP-1,increased the expression of MMP-13 and decreased the expression of p-Erk and p-JNK,and had no effect on p-p38. CONCLUSIONAqueous extract of RC-H might inhibit the activation of tHSC/Cl-6stimulated by TGF-β via phosphorylation of JNK and Erk in MAPK signal pathway.
