黄芪多糖对运动损伤心肌的影响
The Effect of Astragalus Polysaccharide on Sports Injury in Myocardium
【摘要】 目的探讨黄芪多糖对小鼠运动损伤心肌端粒酶表达与血清超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-px)、丙二醛(MDA)含量之间的关系。方法取昆明种小鼠随机分为4组,黄芪多糖大、小剂量组、运动损伤对照组和空白对照组,连续灌胃给药21 d,于末次给药后1 h除空白组外,其他3组小鼠行力竭游泳运动60min,取血测定SOD、GSH-px和MDA的含量,检测骨骼肌形态学变化及心肌端粒酶表达。结果黄芪多糖可以提高运动损伤小鼠血清SOD和GSH-px活性,降低MDA的含量,与运动损伤组比较P<0.01;心肌端粒酶运动损伤组表达最多,空白组最少,黄芪多糖能下调心肌端粒酶表达,与运动损伤组相比P<0.05。镜下可见运动损伤组小鼠骨骼肌纤维炎性细胞浸润增多,肌细胞出现不同程度水肿、变性和坏死;黄芪多糖大、小剂量组骨骼肌细胞数目增加,未发现明显水肿、变性和坏死;心肌端粒酶表达与血清SOD之间呈正相关,与MDA之间呈负相关,与GSH-px相关性较小。结论黄芪多糖通过提高运动损伤小鼠SOD和GSH-px的活性,降低MDA的含量,下调心肌端粒酶表达,起到保护运动损伤的作用。
【Abstract】 Objective To explore the relations among lab mouse sport injury myocardial telomerase expression under astragalus polysaccharide,serum superoxide dismutase(SOD),glutathione peroxidase(GSH-px),and malondialdehyde(MDA).Methods Randomly divide the Kunming mice into 4 groups:APS high-dose group,APS low-dose group,the sport injury control group and the blank control group.Consecutive treatment in stomach,21 days,1 hour after the last dose,except the blank control group,the other 3 groups of mice swim for 60 minutes.Collect the blood sample to measure SOD,GSH-px and MDA content,detect the morphological changes in skeletal muscle and myocardium telomerase expression.All the experimental data are presented as mean ± standard deviation(x±s),statistical processing is tested in F.Results Astragalus polysaccharides can improve sports injuries mice serum SOD and GSH-px activity,decreased MDA content,comparing with the Sports Injury group(P <0.01);the expression of myocardial telomerase is most in sports injury mice group,and least in the blank control group.APS can lower the expression of myocardial telomerase,comparing with the sport injury group(P <0.05).Microscope showed that the increase in sports injuries mice skeletal muscle fiber infiltration of inflammatory cells,muscle cells showed varying degrees of edema,degeneration and necrosis;in Astragalus Polysaccharide high and low dose group,there is an increase in the number of skeletal muscle cells and no obvious edema,degeneration and necrosis are found.Relevant analysis found a positive correlation between the myocardial expression of telomerase and SOD,and a negative correlation between MDA and a small correlation between GSH-px.Conclusions Astragalus polysaccharides can enhance sports injury mice’s SOD and GSH-px activity,decrease MDA content,reduce myocardial expression of telomerase,and plays a role in the protection of sports injuries.
