【摘要】 目的 探讨Twist蛋白在肾肿瘤组织中的表达及与几种多药耐药蛋白表达之间的关系。方法 应用免疫组化SABC技术检测人肾癌组织不同区域的Twist蛋白、多药耐药相关蛋白(multidrug resistance-associated protein,MRP)、P糖蛋白(P-gp)、肺耐药蛋白(lung resistance-related protein,LRP)、拓扑异构酶Ⅱ(topoisomeraseⅡ,TOPOⅡ)的表达。结果 在肿瘤边界区,Twist蛋白、MRP、P-gp、LRP、TOPOⅡ均为阳性表达;而在肿瘤中心区,Twist蛋白和P-gp的表达为阴性,MRP、LRP、TOPOⅡ蛋白仍为阳性表达,但MRP和TOPOⅡ在肿瘤中心区表达强于肿瘤周边区(P <0.01),而LRP在肿瘤中心区的表达弱于肿瘤周边区(P <0.01)。结论Twist基因的表达与肾肿瘤转移、浸润以及耐药性有关。更多还原

【Abstract】 Objective To investigate the relationship between the expressions of Twist protein in renal carcinoma tissue and several drug resistance proteins.Methods The expressions of Twist protein,multidrug resistance-associated protein(MRP),P-glycoprotein(P-gp),lung resistance protein(LRP),and topoisomerase Ⅱ(TOPO Ⅱ) in different zones of human renal carcinoma tissues were detected using streptavidin-biotin-peroxidase complex(SABC) immune histochemistry technique.Results Immunostaining results showed that the expressions of Twist protein,MRP,P-gp,LRP,and TOPO Ⅱ were positive in the boundary zone of renal carcinoma,while in the cancer center zone,the expressions of Twist protein and P-gp were negative.MRP,LRP,and TOPO Ⅱ were still positively expressed in cancer center zone,and the expression levels of MRP and TOPO Ⅱ were higher in cancer center zone than those in boundary zone(P <0.01),while the expression level of LRP was lower than that in boundary zone(P <0.01).Conclusion Twist gene is related to metastasis,infiltration,and drug resistance in renal carcinoma.更多还原
