RAD001对胃癌耐药细胞株SGC7901/DDP的VEGF表达的影响

【摘要】 目的 研究哺乳动物雷帕霉素靶蛋白(mammalian target of rapamycin,mTOR)特异性抑制剂RAD001对胃癌耐药细胞株SGC7901/DDP的血管内皮生长因子(vascular endothelial growth factor,VEGF)表达的影响,以期为临床治疗肿瘤提供新的线索。方法 常规培养胃癌耐药细胞株SGC7901/DDP,RAD001单独或联合顺铂(DDP)干预48 h后,ELISA法测定药物对细胞分泌VEGF蛋白的影响,蛋白免疫印迹法和免疫细胞化学法检测药物作用后细胞中VEGF蛋白的表达情况。结果RAD001单独或联合DDP干预后,SGC7901/DDP细胞分泌的VEGF蛋白减少,呈剂量依赖性;SGC7901/DDP细胞的VEGF蛋白的表达降低。结论 RAD001可以减少胃癌SGC7901/DDP细胞的VEGF蛋白的分泌和表达,与DDP联合用药时作用更明显。

【Abstract】 Objective To investigate the effect of mTOR inhabitor RAD001 in the expression of VEGF in SGC7901/DDP cells.Methods SGC7901/DDP cells were cultured in RPMI 1640 containing 10% FBS,500 μg/L cisplatin(DDP),100 × 10~3 U/L penicillin and 100 mg/L streptomycin.Then the cells were treated with RAD001 alone or in combination with DDP.ELISA was used to detect the secreted VEGF levels of SGC7901/DDP.Western blot assay and immunocytochemistry were used to detect the expression of VEGF.Results After treaded with RAD001 or combination with DDP,the secreted VEGF and expression of VEGF was inhabited in a dose-dependment manner.Conclusion RAD001 can inhibit the secretion and expression of VEGF in SGC7901/DDP cells.If combined with DDP,the inhibition effect will be more obvious.
