白木通基因组DNA提取方法研究
Method for Extracting Genomic DNA from Leaves of Akebia Trifoliata(Thunb.) Koidz. var. Australis(Diels)
【摘要】 以白木通新鲜叶片为材料,采用改良CTAB法提取DNA,并对提取的DNA进行了纯度鉴定和PCR检测。结果表明:改良的CTAB提取法,能够有效去除次生物质对DNA的干扰,提高提取DNA的质量和纯度,适宜白木通叶片DNA的提取。该改良法提取的DNA可用于SRAP分析。
【Abstract】 With fresh leaves of Akebia trifoliata( Thunb.) Koidz. var. Australis( Diels) as materials,the modified CTAB method is adopted to extract DNA. The purity test and the PCR test were conducted to the DNA. The results showed that it can effectively eliminate the interfere of secondary substances to DNA and improve quality and purity of extracted DNA when CATB method was modified. The modified CTAB method is suitable for extracting DNA from fresh leaves of Akebia trifoliata( Thunb.) Koidz. var. Australis( Diels),and the extracted DNA can be used in SRAP analysis.
