黄芪多糖对RAW264.7巨噬细胞源性泡沫细胞胆固醇流出及PPARγ表达的影响
Effects of Astragalus Polysaccharides on cholesterol efflux and the expression of PPARγ in RAW264.7 macrophage-derived foam cells
【摘要】 目的 以RAW264.7巨噬细胞源性泡沫细胞为研究对象,观察不同浓度黄芪多糖(APS)对细胞内胆固醇流出及PPARγ表达的影响,探讨可能作用机制。方法 将RAW264.7细胞诱导分化为泡沫细胞,用油红O染色法鉴定。不同浓度APS作用泡沫细胞48 h后,液体闪烁计数仪检测胆固醇的流出率,并用RTPCR及ELISA测定PPARγ的基因及蛋白表达。结果 ①巨噬细胞经氧化型低密度脂蛋白(ox-LDL)诱导48 h后转化为泡沫细胞。②与对照组相比,不同浓度APS干预组泡沫细胞的胆固醇流出率上升(P<0.01),且在一定浓度范围内呈剂量依赖性。③与对照组相比,不同浓度APS干预组泡沫细胞内PPARγ的基因表达上调(P<0.01),且在一定浓度范围内呈剂量依赖性。④与对照组相比,20、50、100mg/L浓度APS干预组泡沫细胞内PPARγ的蛋白表达增加(P<0.01)。结论 ①经ox-LDL诱导后,巨噬细胞分化为泡沫细胞,细胞内脂质大量增加。②APS促进巨噬细胞内胆固醇流出,可能与上调细胞内PPARγ的表达有关。
【Abstract】 Objective To study the effects of Astragalus Polysaccharides(APS) on cholesterol efflux and the expression of PPARγ in RAW264.7 macrophage-derived foam cells.Methods The mouse RAW264.7 cells were induced into foam cells,then we applied red oil 0 staining technique to appraise the foam cells.The RAW264.7 macrophage-derived foam cells were treated with APS at different concentrations.The cholesterol efflux of foam cells was determined by Liquid Scintillation Counter;the expression of PPARγ mRNA and protein was determined by RT-PCR and ELISA.Results(1)The macrophage cells were differentiated into foam cells after 48hours by ox-LDL.(2)Compared with the control group,treatment of RAW264.7 macrophage-derived foam cells with different concentrations of APS increased the efflux of intracellular cholesterol in a dose-dependent manner(P<0.01).(3)Compared with the control group,treatment of RAW 264.7 macrophage-derived foam cells with different concentrations of APS upregulated the expression of PPAR7 mRNA in RAW264.7 cells in a dose-dependent manner(P<0.01).(4)Compared with the control group,treatment of RAW264.7 macrophage-derived foam cells with20 mg/L,50 mg/L,100 mg/L APS for 24 h resulted in an increase in the expression of PPARγ protein(P<0.01).Conclusion(1)The mouse RAW264.7 cells were differentiated into foam cells after 48 hours by ox-LDL,and the Goncentration of intracellular lipid increased.(2)APS may enhance the efflux of intracellular cholesterol by upregulating PPARγ expression in RAW264.7 cells.
