免疫刺激序列对鼠实验性过敏性结膜炎的作用研究
Effects of immunostimulatory sequence on experimental allergic conjunctivitis
【摘要】 目的构建豚草花粉诱导的小鼠过敏性结膜炎动物模型,观察免疫刺激序列胞嘧啶-磷酸-鸟嘌呤(cytidine-phosphate-guanosine,CpG)DNA对过敏性结膜炎的免疫刺激作用。方法将16只BALB/c小鼠分为CpG DNA实验组和对照组,每组各8只,均于左足部局部注射豚草花粉和氢氧化铝悬浮液进行初次致敏,在注射后第14天右眼结膜囊内滴用豚草花粉溶液进行再次致敏;其中CpG DNA实验组在再次致敏前3d给予CpG DNA滴眼,对照组给予PBS滴眼。观测指标包括症状得分、眼穹隆部结膜嗜酸性粒细胞的数量及结膜IL-12p40 mRNA的表达情况。结果 CpG DNA实验组小鼠仅偶有眼睑或球结膜充血水肿等过敏反应,眼局部反应均较对照组明显减轻(均为P<0.01)。CpG DNA实验组临床症状得分为(5.50±1.60)分,明显低于对照组的(10.88±1.73)分,差异有统计学意义(t=3.560,P=0.003)。CpG DNA实验组小鼠穹隆部结膜组织中嗜酸性粒细胞每高倍视野镜下细胞计数为(16.12±9.31)个,明显少于对照组的(67.25±20.63)个,差异有统计学意义(t=6.39,P<0.01)。CpG DNA实验组结膜组织中IL-12p40 mRNA的表达上调,是对照组IL-12p40 mRNA相对表达量的9.2倍。结论在致敏初始阶段给予CpG DNA能够明显抑制小鼠过敏性结膜炎的过敏反应和晚期炎症反应,其作用可能与CpG DNA诱导IL-12p40过表达有关。
【Abstract】 Objective To construct the animal model of allergic conjunctivitis and observe the effect of immunostimulatory sequence(CpG DNA) on experimental allergic conjunctivits.Methods A total of 16 BALB/c mice were equally divided into CpG DNA group and control group.They were all injected topically with suspension of ragweed pollen and aluminum hydroxide on the left footpad.At 14 days or so,they were all again injected with suspension of ragweed pollen.CpG DNA group was administered topically with conjunctiva 3 days before allergen challenge,while control group was administered with PBS.Multiple parameters including scores of clinical symptoms,expression of eosinophilia in forniceal region of the conjunctiva and expression of IL-12 p40 mRNA in the conjunctiva were measured.Results In CpG DNA group,eye local reaction except individual conjunctival and lid edema and redness relieved obviously(all P<0.01).The clinical total scores in CpG DNA group was significantly less than control group(5.50±1.60 vs 10.88±1.73),and there was significant difference between them(t=3.560,P=0.003).The number of eosinophilia in conjunctiva decreased dramatically in CpG DNA group(16.12±9.31 vs 67.25±20.63),and there was significant difference between them(t=6.39,P<0.01).The expression of IL-12 p40 mRNA in CpG DNA group increased significantly,as much 9.2 times as that of control group.Conclusion Early conjunctival administration of CpG DNA can significantly inhibit allergic responses and late-phase inflammation reaction of mice allergic conjunctivitis.Its effect may relate to the over expression of IL-12 p40 induced by CpG DNA.
