HPLC法测定新疆白喉乌头不同部位氢溴酸高乌甲素的含量

【摘要】 目的比较新疆尼勒克地区白喉乌头不同部位(根、茎、叶)中氢溴酸高乌甲素的含量。方法采用X Bridge C18生物碱专用色谱柱(250mm×4.6mm,5μm),以0.04mol/L醋酸铵-乙腈(63∶37)为流动相等度洗脱,流速0.8mL/min,柱温40℃,检测波长为252nm,进样量10μL高效液相色谱法测定白喉乌头根、茎、叶中氢溴酸高乌甲素含量。结果白喉乌头中氢溴酸高乌甲素在根中含量为0.203 8mg/g,在叶中的含量为0.061 1mg/g,在茎中的含量为0.011 1mg/g。结论白喉乌头根中氢溴酸高乌甲素含量高于叶和茎中,为白喉乌头资源的进一步利用奠定了一定基础。

【Abstract】 Objective The study is to compare hydrobromide lappaconitine content in different parts(root,stem,leaf)of Aconitum leucostomum Woroschby using HPLC method.Methods Use X Bridge C18 Alkaloid chromatographic column(250mm×4.6mm,5μm)and take 0.04mol/L solution of ammonium acetate-acetonitrile as solvent to elute with same flow rate of 0.8ml/min under the column temperature 40℃.The detection wavelength is set as 252nm,and injection volume was 10μL.Results The content of hydrobromide lappaconition in the root of Aconitum leucostomum Woroschis 0.2038mg/g,0.0611mg/g in leaf and 0.0111mg/g in stem.Conclusion The content of hydrobromide lappaconition in Aconitum leucostomum Woroschin root is higher than that is in leaf and stem.The studies lay the foundation for further application of Aconitum leucostomum Woroschresources.
