正常盐饮食状态下醛固酮增多对大鼠主动脉纤维化的作用和机制研究

【摘要】 目的观察大鼠在正常盐饮食情况下,醛固酮增多对SD大鼠主动脉的影响、血清炎症因子[白细胞介素-6(IL-6)和单核细胞趋化蛋白-1(MCP-1)]表达和大鼠主动脉组织中与血清炎症因子相对应的受体表达的变化。方法实验动物随机分正常组和醛固酮组,每组8只,醛固酮组采用醛固酮皮下注射(20μg/d),常规饲养6周后,用Masson染色血管胶原沉积情况,用放射免疫法检测血清炎症因子含量,用免疫组织化学法检测血管组织炎症因子受体表达。结果 Masson染色可见醛固酮组大鼠主动脉有大量胶原沉积,醛固酮组大鼠血清炎症因子(IL-6、MCP-1)较正常组明显上调[分别为(187.00±6.58)比(150.90±5.12)ng/L,(83.06±2.79)比(70.73±2.02)ng/L,均为P<0.01];与正常组相比,醛固酮组大鼠主动脉组织中的炎症因子相应受体(IL-6受体和MCP-1受体)的表达亦同步上调。结论醛固酮组大鼠主动脉胶原明显沉积,血清炎症因子和主动脉相应受体表达上调,提示醛固酮不仅是促纤维化因子,而且是促炎症因子。

【Abstract】 Objective To investigate the role and possible mechanism of excess aldosterone on the aorta of rats with the absence of high salt diet. Methods SD rats were randomly divided into normal group( n = 8) and aldosterone group( n = 8). Rats in aldosterone group were infused subcutaneously with aldosterone( 20 ug / d) for 6 weeks. The expression of collagen was determined by Masson staining.Inflammatory factor contents in serum were determined by radioimmunoassay, and the expressions of corresponding receptors were determined by immunohistochemistry. Results Masson staining showed that there was massive collagen deposit in aortic wall of aldosterone rats. The contents of inflammatory factors in serum in aldosterone group were obviously raised compared with those in the normal group [IL-6( 187. 00 ±6. 58) vs.( 150. 90 ± 5. 12) ng / L,MCP-1( 83. 06 ± 2. 79) vs.( 70. 73 ± 2. 02) ng / L,P < 0. 01] and the expressions of corresponding receptors in aldosterone group were significantly up-regulated at the same time.Conclusions Inflammatory factors and the corresponding receptors are significantly up-regulated in aldosterone group,which suggests that aldosterone is not only the factor for fibrosis,but also the proinflammatory factor.
