小干扰RNA沉默高迁移率族蛋白1对胃癌细胞生物学行为的影响
Effect of HMGBl-si RNA on biological behavior of human gastric cancer
【摘要】 目的靶向高迁移率族蛋白1(high mobility group protein,HMGBl)的小干扰RNA(si RNA)转染胃癌HGC-27细胞株抑制HMGBl基因表达,探讨其对胃癌细胞增殖、侵袭和迁移的影响。方法将靶向HMGB1的si RNA在脂质体介导下转染胃癌HGC-27细胞,Western blot检测转染后细胞HMGB1蛋白表达变化,通过MTT实验检测转染后细胞的增殖活性,细胞划痕实验检测细胞侵袭和迁移能力。结果空白转染组、control-si RNA组和HMGB1干扰组HMGB1蛋白相对表达水平分别为(0.940±0.059)、(0.927±0.051)和(0.393±0.033),HMGB1干扰组蛋白表达水平明显下降,差异有统计学意义(P<0.05)。MTT和细胞划痕实验显示,与对照组相比,HMGB1干扰组细胞增殖缓慢,细胞迁移率明显降低,差异有统计学意义(P<0.05)。结论应用靶向si RNA沉默胃癌HGC-27细胞HMGB1基因表达可抑制其增殖、侵袭和迁移能力,具有抗肿瘤作用,HMGBl有望成为胃癌治疗的一个潜在靶点。
【Abstract】 Objective To investigate downregulation of HMGB1 by si RNA and its effect on cell proliferation, invasion and migration of huaman gastric cancer cell line HGC-27. Methods HMGBl-si RNA was transfected into HGC-27 cells by lipofectamine TM 2000. Cells transfected with control-si RNA and vehicle were used as control groups. HMGB1 expression was detected by Western blot. Cell proliferation was measured by MTT assay. Cell migration was evaluated through the cell scratch method. Results The expression level of HMGB1 in vehicle group, control-si RNA group and HMGBl-si RNA group were(0.940 ±0.059),(0.927 ±0.051) and(0.393 ±0.033) respectively. Compared with the two control groups, the expression of HMGB1 in HMGBl-si RNA group was significantly decreased(P<0.05). Cell proliferation in the HMGBl-si RNA group was inhibited significantly and cell migration significantly decreased compared with those in the two control groups(P <0.05). Conclusions The expression of HMGB1 can be reduced significantly by HMGB1-si RNA. HMGB1 downregulation can suppress cell proliferation and migration of human gastric cancer, which can provide a potential target therapy for gastric cancer.
