5-Fu联合乌司他丁对结肠癌HT-29细胞裸鼠移植瘤的抑制作用

【摘要】 目的观察5-氟尿嘧啶(5-fluorouracil,5-Fu)和乌司他丁联合应用对结肠癌的抑制作用。方法构建结肠癌HT-29裸鼠皮下移植瘤模型。腹腔药物注射,分为4组:对照组(A组)、单独乌司他丁组(B组)、单独5-Fu组(C组)、5-Fu和乌司他丁联用组(D组),观察二者单独及联合作用对结肠癌移植瘤生长的作用。结果给药21天时,A、B两组移植瘤体积无明显差异(P>0.05),C、D组移植瘤体积均小于A组(P<0.05),其中D组移植瘤体积低于C组(P<0.05)。四组小鼠用药前体重无显著差异(P>0.05),用药后A、B两组移植瘤重及非肿瘤体重无明显差异(P>0.05),C、D两组移植瘤重均低于A组(P<0.05),非肿瘤体重均高于A组(P<0.05),且D组移植瘤重低于C组(P<0.05),非肿瘤体重高于C组(P<0.05)。结论 5-Fu和乌司他丁联合应用,对结肠癌移植瘤抑制作用增强,可改善化疗后的生存状态。

【Abstract】 Objective To observe the inhibitory effect of 5- fluorouracil(5-Fu) combined with ulinastatin on xenograft of colon caner HT-29 in nude mice. Methods After Colon cancer HT-29 xenograft models in nude mice were established, intraperitoneal injection of saline(group A), ulinastatin(group B),5-Fu(group C), and 5-Fu combined with ulinastatin(group D) was administered. The volume of xenografted tumor, tumor weight and non-tumor weight were compared among the different groups. Results On the 21st day after intraperitoneal injection, there was no difference in tumor volume between the group A and group B(P>0.05), the tumor volume was significantly decreased in the group C and D compared with the group A(P<0.05), furthermore the tumor volume in the group D was less than that in the group C. There was no difference in weight before intraperitoneal injection among the four groups. There were no differences in tumor weight and non-tumor weight between the group A and B. The tumor weight was significantly lower, nontumor weight higher in the group C and D respectively than those in the group A. The tumor weight was lower, non-tumor weight higher in the group D than those in the group C. Conclusion Combined use of 5-Fu and ulinastatin can enhance the inhibition of colon cancer xenografts, and improve survival status after chemotherapy.
