AAV2/9介导的PD-L1保护移植物的实验研究
EXPERIMENTAL STUDY ON ADENO-ASSOCIATED VIRUS 2/9 MEDIATED PD-L1 GENE TRANSFER TO THE GRAFT PROTECTING ALLO-REJECTION
【摘要】 目的探讨腺相关病毒介导的PD-L1在移植排斥反应中的作用。方法用含AAV2/9-PD-L1或LacZ的UW液灌注供体心脏后,移植到受体大鼠体内,观察移植物的转染效率和存活时间。结果AAV2/9可以高效地介导外源性基因转染移植物,AAV2/9-PD-L1转染移植物后虽能延长移植物的存活时间,但和对照组比较无统计学意义。结论 AAV2/9是一种高效安全的病毒载体,PD-L1局部表达抑制排斥反应的机制还需进一步研究。
【Abstract】 Objective To investigated the effect of Adeno-associated viral vectors 2/9(AAV2/9)mediated program death ligand 1(PD-L1) gene transfer to myocardium on cardiac allograft survival in a rat heart transplantation model. Methods Animals were divided into three groups: GroupPD-L1:0.3 ml UW solution containing AAV2/9-PD-L1(5×1012 vector genome,vg) was infused into Brown Norway(BN)rat hearts prior to be transplanted into Lewis rats.In control group,null AAV2/9 viral vectors and UW solution were applied.To evaluate the gene transfection efficiency,additional group of AAV2/9-LacZ were used in iso-transplantation group. Results Efficient transfection was demonstrated by application of AAV2/9-LacZ in iso-graft.Successful transfection of AAV2/9-PD-L1 prolonged the allograft survival but without statistical significance. Conclusion Gene transfer of PD-L1 to graft could protect the allograft from the allo-rejection.The mechanism should be further studied.
