胚胎癌F9细胞中miR-302与Oct4表达相关性研究

【摘要】 目的探讨F9细胞中miR-302是否是Oct4的靶基因之一。方法采用荧光半定量real-timePCR动态检测维甲酸(RA)诱导胚胎癌F9细胞分化过程中及Oct4的RNA干扰后F9细胞中Oct4及miR-302的表达量的变化。结果 (1)RA诱导F9细胞分化过程中,Oct4表达量在诱导24h后下降了44%,而48h后下降了71%;与Oct4表达模式相似,miR-302的表达在诱导24h后下降了37%,而48h后下降了63%。(2)Oct4RNA干扰后Oct4表达下降了56%,miR-302的表达下降了38%。结论 F9细胞中Oct4及miR-302的表达具有相关性,miR302可能是Oct4的靶基因之一。

【Abstract】 OBJECTIVE The aim of this study was to investigate whether miR-302 is a target gene of Oct4 in the F9 mouse embryonic carcinoma cells or not. METHODS The expression of Oct4 and miR-302 in F9 cells after retinoid acid (RA) treatment for various periods and after Oct4 siRNA interference were analyzed by real-time PCR. RESULTS The levels Oct4 decreased 44% by 24h and 71% by 48h after RA treatment. Similarly, miR-302 expression decreased 37% by 24h and 63% by 48h after treatment. After siRNA-mediated knockdown of Oct4, Oct4 decreased 56% and miR-302 expression decreased 38%. CONCLUSION These data indicat that Oct4 and miR-302 are coordinately expressed in F9 cells, and the miR-302 may be regulated by Oct4.
