山西五台山不同海拔中国沙棘居群的遗传变异

ISSR analysis of genetic diversity of H rhamnoides ssp sinensis at different altitudes at Wutai Mountain in Shanxi Province

【摘要】 利用ISSR分子标记分析了山西五台山不同海拔中国沙棘居群的遗传多样性和遗传分化.用11条引物对3个中国沙棘居群共60个样品进行扩增,共检测到170个位点,其中多态位点162个,多态位点百分率(PPB)为95.29%.五台山中国沙棘总的Shannons信息指数(I)为0.455 6,Neis指数(h)为0.301 1,表明亚种水平的遗传多样性较高.五台山中国沙棘居群水平也具有很高的遗传多样性,PPB平均为62.55%,I平均为0.323 5,h平均为0.203 0.五台山高海拔的中国沙棘居群S15和S16较低海拔居群S14遗传多样性丰富.POPGENE分析表明,五台山中国沙棘居群间存在较高的遗传分化(Gst=0.316 1),在总的遗传变异中31.61%的变异存在于居群间,居群间的基因流仅为1.081 8,限制的基因流可能是居群间存在较高遗传分化的主要原因,较高的居群间的遗传分化暗示在资源保护和利用中应考虑对不同地方居群的保护和利用.

【Abstract】 In this paper,Hippophae rhamnoides ssp sinensis populations are collected from different altitudes at Wutai Mountain in Shanxi Province,and their genetic diversity and genetic differentiation are analysed by using the inter simple sequence repeat(ISSR) molecular technique.ISSR amplification of 60 individuals from 3 populations is conducted with 11 primers,and 162 polymorphic loci are detected from total 170 loci,with the percentage of polymorphic bands(PPB) being 95.29%.The Shannon’s information index(I) and Nei’s gene diversity(h) of H rhamnoides ssp sinensis from Wutai Mountain at species level are 0.455 6 and 0.301 1 respectively,indicating a high genetic diversity of H rhamnoides ssp sinensis at species level.High genetic diversity of H rhamnoides ssp sinensis at population level is also detected,with the mean values of PPB,I and h being 62.55%,0.323 5 and 0.203 0 respectively.Population S15 and S16,which distributed at altitude of 203 3 m and 162 4 m respectively of Wutai Mountain,have higher genetic diversity than S14 at 121 4 m.POPGENE analysis shows the genetic differentiation coefficient(Gst) among the populations being 0.316 1.That mean among population component account for 31.61% of the total variation,the gene flow among the populations is only 1.081 8.The high genetic differentiation between populations implies the protection and utilization of resources should be considered in different areas’ populations.
