西葫芦胚囊植株生殖特性
Reproductive Characteristics of Embryo Sac Plants in Summer Squash(Cucurbita pepo L.)
【摘要】 考察了经未受精胚珠离体诱导获得的西葫芦再生胚囊植株及其3个自交世代的生殖特性。单倍体胚囊植株高度不育,无法获得自交后代。绝大多数双单倍体(DH)胚囊植株及其3个自交世代的花器特征、开花进程等与二倍体供体一致。DH植株当代的花粉萌发率为15.52%~42.67%,花粉量偏少,自交单果结籽数明显少于二倍体供体。DH植株3个自交世代的花粉萌发率为17.62%~42.70%,均低于二倍体供体,各自交株系的花粉生活力在3个自交世代间保持稳定,但花粉量仍偏少。交叉授粉验证结果显示,DH植株当代及其3个自交世代的雌性生殖能力接近二倍体供体,雄性生殖能力明显低于二倍体供体,但未呈现持续下降趋势。DH植株3个自交世代的种子发芽率均在92%以上,自交种子具可稔性。
【Abstract】 Reproductive characteristics of embryo sac plants derived from unfertilized ovule in summer squash through in vitro culture and its three self-bred generations were investigated.The embryo sac plants included haploid and double haploid(DH) plants.Haploids had drastic reduction in plant height,poor viability and male and female sterility.DH plants and its three self-bred generations were similar in floral organ characteristics and flowering habit to general diploid except individual plant with abnormal floral organ.Pollen germination rates of DH plants ranged from 15.52%-42.67%,but had less pollen.Seeds per fruit were obviously less than diploid.Pollen germination rates of three self-bred progenies ranged from 17.62%-42.70%,and remained stable between three self-bred progenies,but were lower than those of diploid,and still had less pollen.Female viability of DH plants and its three self-bred generations was closed to that of diploid,and maintained stability between three self-bred generations.Male viability was significantly lower than diploid but didn’t present downtrend,and had also stability between three self-bred generations.Seeds of three self-bred generations had normal self-bred fertility with seed germination rates above 92%.
