等位基因特异扩增检测水稻抗稻瘟病基因Pi-ta
Detection of Rice Pi-ta Gene by Allele-specific PCR
【摘要】 为明确陕西省主要水稻种质资源中抗稻瘟病基因Pi-ta的存在状况,为稻瘟病抗性育种提供依据。利用在抗稻瘟病基因Pi-ta区域设计的四引物扩增受阻突变体系PCR方法,对陕西省2010-2012年水稻区域试验的327份材料进行Pi-ta基因位点检测。结果表明,4份材料为抗性基因纯合体,40份材料为抗性基因杂合体,276份材料在Pi-ta位点为感性基因纯合体,7份材料未检出条带。检测结果真实可靠,可以作为检测水稻Pi-ta基因位点的有效方法。
【Abstract】 The study aimed to determine the existence of Pi-ta gene for rice blast resistance in the rice germplasm in Shaanxi Province,and provide a theoretical basis for the breeding of rice blast resistance.The distribution of this gene among 327 rice germplasm from Shaanxi rice regional test in 2010,2011 and 2012 was analyzed by using Tetra-primer ARMS.The results showed that there were 4 lines with homozygous genotype of Pi-ta resistance gene,40 lines with heterozygous genotype,276 lines with susceptible homozygous genotype,and 7 lines could not detect any bands.The results are reliable and the methods can be effectively used in the detection of Pi-ta gene in rice.
