高效阴离子交换-脉冲安培检测壳寡糖在体外非翻转肠囊模型中的吸收

【摘要】 建立高效阴离子交换色谱结合脉冲安培检测器(HPAEC-PAD)检测非翻转肠囊中壳寡糖吸收的方法。结果表明,以RCX-10阴离子交换柱为分离柱,通过NaOH溶液的梯度淋洗,壳一糖~壳六糖的检出限为0.8~4.0mg/L(进样体积100μL,峰面积定量),该方法的相对标准偏差为1.3%~6.6%。体外非翻转大鼠肠囊模型中,壳二糖、壳三糖以及壳四糖在十二指肠、空肠以及回肠中均有良好的吸收(Papp>1×10-5 cm/s),在结肠中仅能部分吸收(Papp<3×10-6 cm/s)。壳寡糖吸收速率随聚合度的升高而降低,壳寡糖吸收符合一级吸收动力学特征,推测壳寡糖在体内的吸收为被动过程。

【Abstract】 A method for determination of chitooligosaccharides(COS) in non-everted intestinal sacs model by high performance anion-exchange chromatography with pulsed amperometric detection(HPAEC-PAD) was established.The chromatographic separation was established on a Hamilton RCX-10 anion exchange column using NaOH gradient elution.Detection limits of 0.8-4.0 mg/L(100 μL injection volume) were achieved under optimized conditions.RSD of the peak area for COS was 1.3%-6.6%.The results demonstrated chitobiose,chitotriose and chitotetraose could be well absorbed in duodenum,jejunum and ileum with high apparent permeability coefficients(Papp>1×10-5cm/s),and the absorption of COS in colon was low(Papp<3×10-6cm/s) in non-everted intestinal sacs in vitro.It was also proven that as polymerization degree of COS increased,the absorption rate decreased.The mechanism of COS absorption was in a passive absorption manner and followed the first-order absorption kinetics.
