解磷、解钾根际促生菌的筛选与鉴定

【摘要】 试验从樱桃根围采集根际土壤样品,采用MRS培养基和改良麦氏培养基,分别在28℃、37℃条件下培养2~3d,进行分离,利用系列稀释涂布平板筛选出分离物DY-3和DY-4。根据菌体形态、菌落形态观察和生理生化试验结果,结合18S或16SrDNA序列的测定,将DY-3鉴定为酿酒酵母菌(Saccharomyces cerevisiae),DY-4鉴定为干酪乳杆菌(Lactobacillus casei)。测其解磷解钾能力,酿酒酵母菌DY-3有较好的解钾能力,干酪乳杆菌DY-4有较好的解磷能力。因此,该研究筛选的2菌株可望进一步研究开发成为微生物肥料的生产菌种。

【Abstract】 The aim of the present study was to provide the excellent strain resources for the production of bio-fertilizer. Therefore,isolates were isolated fromCerasus pseudocerasus rhizospheric soil at 28℃,37℃for 2-3d.Two isolates named DY-3and DY-4were screened using modified MacConkey medium and MRS medium,respectively.According to the results of morphologic characteristics,physiological biochemical properties and phylogenetic analysis o f 16SrDNA genes,DY-3was identified as Saccharomyces cerevisiae,as well as DY-4identified as Lactobacillus casei.And then,the phosphate-dissolving and potassium~dissolving ability of DY-3and DY-4was detected.Result showed that DY-3has the ability of potassium-dissolving,and DY-4has the ability of phosphate-dissolving.As a result,the strains of DY-3and DY-4are possible to be further developed as excellent strains for microbial fertilizer production.
