红腺忍冬花中绿原酸提取工艺研究

【摘要】 本文对红腺忍冬花中绿原酸的提取工艺进行研究,优化提取条件。在提取过程中通过单因素实验,分析了乙醇浓度、料液比、溶剂pH值、提取时间及提取次数等五个因素对提取率的影响,在此基础通过四因素三水平正交实验对工艺条件进行优化,以绿原酸的提取率为指标,实验结果表明在最佳吸收波长329 nm处,红腺忍冬花中绿原酸提取最佳工艺条件为:乙醇浓度为70%、料液比为1∶10、提取时间为2 h、提取次数为2。在最佳工艺条件下,绿原酸提取率为4.85%。以绿原酸为对照品,用紫外可见分光光度法测定红腺忍冬花提取物中绿原酸含量,方法简单,稳定,准确度较高。

【Abstract】 The extraction method of chlorogenic acid from flower of Lonicera hypoglauca Miq. was investigated and optimized. Five influencing factors including aqueous ethanol concentration,ratio of material / solvent,pH value of solvent,extraction duration and times of extraction were studied using single factor analysis method. In addition,the L9( 34) orthogonal tests were designed and used to optimize the extraction conditions of the chlorogenic acid from L. hypoglauca flowers. The results showed that the optimal extraction conditions was followed as: 70% ethanol as extraction solvent,1∶ 10 as ratio of material / solvent,extracting 2 h,4 as pH value of solvent and with 2 times extraction. Under the optimal extraction condition,the extraction rate of chlorogenic acid was up to 4. 85%. The content of chlorogenic acid in L. hypoglauca flowers was determined using ultraviolet-visual spectrophotometry method,which was proved to be practical,stable and accurate.
