没食子酸酯对神经细胞的抗氧化保护作用研究
Effects of Gallate Esters on Antioxidation of Nerve Cells
【摘要】 神经细胞的氧化损伤是导致各种神经退行性疾病的重要原因之一,为探讨不同的没食子酸酯化合物对神经细胞抗氧化保护作用,本文比较了不同没食子酸酯化合物的细胞毒性,分析了不同浓度的没食子酸酯化合物在过氧化氢氧化胁迫下对神经细胞生长的影响,结果表明没食子酸乙酯、没食子酸正丙酯、没食子酸异戊酯都能有效提高经过氧化氢损伤的SH-SY5Y细胞的存活率,显示对神经细胞的氧化应激损伤具有一定保护作用。特别是从三峡特有濒危植物疏花水柏枝中分离到一种没食子酸甲酯类衍生物,3,4-二甲氧基-5-羟基苯甲酸甲酯,表现出比没食子酸正丙酯更好的抗氧化保护作用,说明其可以作为一种潜在的抗神经退行性疾病药物进行开发和利用。
【Abstract】 Oxidative damage of nerve cells is one of the important causes leading to neurodegenerative diseases. In this study,the protective effect of gallate esters on nerve cells against oxidation was investigated. The cytotoxicity of gallate esters was also studied. The results indicated that ethyl gallate,propyl gallate and isoamyl gallate can effectively improve SH-SY5Y cell’s survival rate under hydrogen peroxide stress,showing a degree of protective activity to the oxidative injury of nerve cells. Especially,methyl 3-hydroxy-4,5-dimethoxybenzoate,a derivative of met4hyl gallate isolated from an endangered plant Myricaria laxiflora endemically grown in the Three Gorges Zone,showed better antioxidant protective effect than propyl gallate,indicating that these gallate esters can be developed as potential drugs for neurodegenerative disease in the future.
