【摘要】目的:研究CCCTC位点结合蛋白(CTCF)对Igκ基因重排的影响。方法:采用RNA干扰技术特异性地下调CTCF的表达,然后利用脂多糖(LPS)诱导祖B细胞(pro-B cell)38B9的分化。利用PCR来定量分析不同的Vκ基因hf24和Vκ21G的重排情况。结果:CTCF在38B9细胞中表达下调后hf24和Vκ21G的重排都有所上升。结论:在pro-B细胞中CTCF对Igκ基因的重排起抑制作用。更多还原

【Abstract】 Objective: To investigate the role of CTCF in the rearrangement of Igκ gene in pro-B cells.Methods: shRNA was used to knockdown CTCF in 38B9 cells.PCR was used to examine the rearrangement of hf24 and Vκ21G.Results: In 38B9 cells,rearrangement of hf24 and Vκ21Gto J region increased after CTCF knockdown.Conclusion: CTCF represses the rearrangement of Igκ gene in pro-B cells.更多还原
