【摘要】 目的 探讨B细胞非霍奇金淋巴瘤(B cell Non-Hodgkin’s Lymphoma,B-NHL)患者骨髓单个核细胞(BMMC)表面CD40、CD40L的表达情况及变化规律,分析CID40、CID40l表达与淋巴瘤发病、侵袭性及预后的关系及意义。方法 采集20例初诊B-NHL伴有骨髓侵犯的患者(实验组)的骨髓标本及12例健康者(对照组)的骨髓标本,分离单个核细胞,采用直接荧光抗体标记,应用流式细胞仪检测细胞表面CD40、CD40L的表达情况。相关实验结果进行统计学比较。结果(1)B-NHL中CD40阳性率(45.78±3.82)%显著高于正常对照组(13.99±2.20)%,P<0.01;B-NHL中CD40L阳性率(3.44±0.88)%显著低于正常对照组(10.64±1.03)%,P<0.01;(2)B-NHL伴有骨髓侵犯的患者CD40、CD40L阳性率与病理类型、LDH值、全身症状呈显著相关,P<0.05;(3)B-NHL伴有骨髓侵犯的患者中CD40与CD40L表达呈负相关,r=-0.47,P<0.05。结论(1)CID40、CD40L阳性率与病理类型、LDH值、全身症状密切相关,其可能与B-NHL的侵袭性及预后有关;(2)B-NHL患者中CD40表达的增高、CD40L表达的减少可能是影响其发病的因素之一;(3)B-NHL患者中CD40L表达的减少可能与肿瘤免疫逃逸机制有关。更多还原

【Abstract】 Objective To explore the expressions and changing regularity of CD40,CD40 Lin surface of bone marrow mononuclear cell(BMMC)of patients with B- cell Non- Hodgkin’s Lymphoma.To study the relationship between the expressions of CD40,CD40L and the pathogenesis.infestation.prognosis,clinical implication of patients with Lymphoma.Methods Twenty bone marrow specimens of B- NHL(experimental group) and twelve healthy bone marrow specimens(contiol group) were collected.To separate mononuclear cell,we used direct fluorescent antibody to mark cells,the expressions of CD40 and CD40L were determined by flow cytometry.Statistical study was done between two groups.Results(1) CD40 positive overexpression rate in B- NHL samples(45.78 ±3.82)%was higher than that in normal samples(13.99 ±2.20)%(P<0.01);CD40L positive overexpression rate in B- NHL samples(3.44 ±0.88)%was lower than that in normal samples(10.64 ±1.03)%(P<0.01).(2)There were significant correlations between the positive overexpression rates of CD40,CD40L and pathology category,lactic dehydrogenase(LDH)levels,the whole body symptoms in B-NHL patients who suffered from bone marrow involvment(P <0.05).(3)There were negative correlations between CD40 and CD40L expressions in B- NHL patients who suffered from bone marrow involvment(r =- 0.47,P < 0.05).Conclusions(1)There are close correlations between CD40、CD40L and pathology category,LDH levels and the whole body symptoms,which can likely be correlative in judging the B-NHL prognosis and infestation.(2)High CD40 expressions and low CD40L expressions may probably be one of etiologic factors of B-NHL.(3)Low CD40L expressions may probably be related with the mechanism of tumor escape.更多还原
