大潮气量机械通气对大鼠肺组织核内受体Nr1d1表达的影响

【摘要】 目的:观察大潮气量机械通气对大鼠肺组织中Nr1d1表达的影响及其作用机制。方法:24只SD大鼠随机分成3组(F组、L组、H组),每组8只,F组为自由呼吸组,L组给予10 mL/kg小潮气量,H组给予40 mL/kg大潮气量行机械通气,通气过程中给予维库溴铵抑制呼吸,通气2 h。通气结束24 h后,取大鼠肺组织检测各组湿干重比值及Nr1d1 mRNA和蛋白的表达。结果:与F组相比,H组W/D值升高而Nr1d1mRNA及其蛋白表达水平均降低。结论:Nr1d1在机械通气肺损伤中可能存在一定作用,其机制可能与肺组织生物节律紊乱和Nr1d1对炎症反应的调控有关。

【Abstract】 Objective To observe the effect of large tide volume mechanical ventilation on the expression of nuclear receptor Nr1d1 in rat lung tissue and the mechanism of action of this ventilation. Methods 24 SD rats were randomly divided into three groups(groups F,L,and H),eight for each group. Group F remained self- breathing,group L received small tidal volume mechanical ventilation of 10 ml / kg,and group H received large tide volume mechanical ventilation of 40 ml / kg. Vecuronium was administered during mechanical ventilation to control breathing,and ventilation lasted for 2 hours. The W / D value and expressions of Nr1d1 mRNA and protein in rat lung tissues were detected 24 hours after ventilation. Results As compared with group F,the W / D value was elevated while expression levels of Nr1d1 mRNA and protein were reduced. Conclusions Nr1d1 may play a role in ventilator-induced lung injury,whose mechanism may be related to distrubed biological rhythm of the lung tissues and to Nr1d1 regulation of inflammatory reaction.
