灵杆菌多糖对内毒素诱导小鼠肺损伤的保护作用
The protective role of hemarisin in endotoxin-induced pulmonary damage in mice
【摘要】 目的:探讨灵杆菌多糖对内毒素(LPS)所致小鼠肺损伤的保护作用。方法:提前0.5 h给小鼠腹腔注射灵杆菌多糖,然后尾静脉注射LPS(30 mg/kg),3 h后ELISA法测定肺组织白介素-1β(IL-1β)、白介素-6(IL-6)、肿瘤坏死因子-α(TNF-α)的含量;6 h后测量肺湿重干重比(W/D)、髓过氧化物酶(MPO)活力、丙二醛(MDA)含量。结果:灵杆菌多糖(100 U/kg)可部分抑制由LPS引起的IL-1β、IL-6、TNF-α的增加(P<0.05),降低肺W/D(P<0.05);剂量为100、50 U/kg时,可部分抑制MPO活性(P<0.05)及MDA含量(P<0.01)的增加。结论:灵杆菌多糖预防性给药对内毒素所致小鼠肺损伤有一定的保护作用。
【Abstract】 Objective To investigate the protective role of hemarisin(lipopolysaccharides from Bacillus prodigiosus) in endotoxin(LPS)-induced pulmonary damage in mice. Methods 0.5 hours after intraperitoneal injections of hemarisin, the mice received LPS of 30 mg / kg via the tail vein. 3 hours after injections of LPS, levels of interleukin-1β(IL-1β), interleukin-6(IL-6), and tumor necrosis factor α(TNF-α) in the lung tissues were detected by ELISA. 6 hours later, the lung wet weight and dry weight ratio(W / D), myeloperoxidase(MPO)activity, and malondialdehyde(MDA) content were determined. Results Hemarisin of 100 U / kg partially inhibited a LPS-induced increase in IL-1β, IL-6, and TNF-α(P < 0.05) and decreased the W / D ratio(P < 0.05).Hemarisin at a dose of 100 or 50 U / kg partially suppressed MPO activity(P < 0.05) and an elevation in MDA content(P < 0.01). Conclusions Prophylactic administration of hemarisin plays a certain role in the protection of endotoxin-induced pulmonary damage in mice.
