舌鳞癌组织中HIF-1α和VEGF表达及与肿瘤血管生成的关系
Expressions of hypoxia-inducible factor-1α and vascular endothelial growth factor in human squamous cell carcinoma of tongue and its correlation with angiogenesis
【摘要】 目的:探讨缺氧诱导因子-1α(HIF-1α)和血管内皮生长因子(VEGF)在舌鳞癌组织中的表达及与微血管密度之间的关系。方法:选择48例舌鳞癌标本,应用免疫组织化学染色方法,检测HIF-1α、VEGF和CD34在舌鳞癌中的表达。对他们之间的相关性进行统计学分析。结果:HIF-1α与VEGF在舌鳞癌组织中阳性均呈高表达,分别为70.8%、64.6%;显著高于正常口腔黏膜阳性表达。舌鳞癌中HIF-1α与VEGF表达与其病理分级、临床分期及淋巴结转移呈相关性(P<0.05)。舌鳞癌中HIF-1α与VEGF表达呈明显正相关性(r=0.554,P<0.01)。HIF-1α和VEGF同为阳性表达的舌鳞癌组织中有较高的微血管密度(t=6.758,P<0.01)。结论:HIF-1α与VEGF在舌鳞癌中的表达显著增高,促进舌癌血管生成,提示在舌鳞癌的发生发展过程中发挥了重要作用。它们的测定可作为判断舌鳞癌恶性潜能的重要生物指标。
【Abstract】 Objective To study the correlation between the expression of hypoxia-inducible factor-1α(HIF1α) and vascular endothelial growth factor(VEGF) and microvessel densitity(MVD) in human squamous cell carcinoma of tongue(SCCT).Methods A total of 48 tongue cancer samples was tested by immunohistochemical method for HIF-1α、VEGF and CD34,and their correlationship were analyzed.Results The positive rates of HIF1α and VEGF in SCCT were 70.8% and 64.6%,respectively,which are significantly higher than those in normal oral mucosa.The positive expression of HIF-1α and VEGF were correlated with pathological grade,TNM stage and lymph node metastasis(P < 0.05).There were positive correlations between the expression of HIF-1α and VEGF(r = 0.554,P < 0.01).SCCT with positive HIF-1αand VEGF expression have higher microvessel density(t = 6.758,P < 0.01).Conclusion The higher expression of both HIF-1α and VEGF can improve the neovascularization of SCCT,which are closely correlated to biological action of SCCT,and play an important role in that development.They can be considered as biological indicators of malignant potential in SCCT.
