三邻甲苯基磷酸酯对人成神经瘤SH-SY5Y细胞内PKC活性及转位的影响
Effect of Tri-ortho-cresyl Phosphate on Activity and Translocation of Protein Kinase C in SH-SY5Y Human Neuroblastoma Cells
【摘要】 目的研究三邻甲苯基磷酸酯(TOCP)对人成神经瘤SH-SY5Y细胞蛋白激酶C(PKC)活性和转位的影响。方法以TOCP(0、0.25、0.5、1.0 mmol/L)对人成神经瘤细胞系SH-SY5Y细胞进行染毒,染毒时间为12、24和48 h,用PKC活性检测试剂盒检测PKC的活性,间接免疫荧光术检测PKC在细胞内的定位,荧光倒置显微镜观察结果。结果 1.0 mmol/L TOCP作用SH-SY5Y细胞24 h,PKC的活性最强,并能导致PKC由胞质向胞膜转位。结论 TOCP作用SH-SY5Y细胞后,能激活PKC,增加膜转位。
【Abstract】 Objective To study the effect of tri-ortho-cresyl phosphate(TOCP) on the activity and translocation of protein kinase C(PKC) in SH-SY5Y human neuroblastoma cells.Methods SH-SY5Y cells were exposed to TOCP at concentrations of 0,0.25,0.5 and 1.0 mmol/L for 12,24 or 48 hours.The PKC activity was detected by PKC kit.The membrane translocation of PKC in cells was determined by indirect immunofluorescence and was observed by inverted fluorescent microscope.Results The PKC in the SH-SY5Y cells treated by TOCP at 1.0 mmol/L for 24 h was the most active,and the translocation of PKC from cytoplasm to membrane was induced.Conclusions TOCP can activate PKC and induce PKC membrane translocation in SH-SY5Y cells.
