生物膜变形链球菌对反义寡核苷酸的摄取率检测

【摘要】 目的:探讨阳离子聚合物So-fast提高生物膜中变形链球菌对反义寡核苷酸摄取率的效果。方法:制备变形链球国际标准株GS5生物膜,在无载体和用阳离子聚合物作载体So-fastTM情况下用变形链球菌gtfB基因的反义脱氧寡核苷酸PS-ODN转化生物膜中变形链球菌,采用流式细胞仪检测生物膜中变形链球菌细菌对PS-ODN的摄取效率。结果:生物膜中的变形链球菌对裸反义寡核苷酸的摄取效率为16%,采用So-fastTM作为转化载体,摄取效率为71.7%。结论:阳离子聚合物可促进反义寡核苷酸通过变形链球菌细胞膜,使其摄取效率明显提高。

【Abstract】 Objective:To study the effect of transfection reagent So-fast in the transfection of PS-ODN into S.mutans in biofilm.Methods: Biofilm of S.mutans GS5 was prepared.Transfection of gtfB phosphorothioate antisense oligonucleotide(PS-ODN) into S.mutans GS5 was conducted with or without transfection reagent So-fastTM.The rate of S.mutans uptaking PS-ODN and PS-ODN/So-fastTM compound was measured by flow cytometry.Results: The rate of S.mutans uptaking PS-ODN in biofilm was 16%.The rate of uptaking PS-ODN/So-fastTM compound in biofilm was 71.4%.Conclusion: Cationic polymer So-fast may increase the uptaking rate of PS-ODN into S.mutans.
