乳杆菌代谢产物诱导人舌鳞癌细胞的凋亡作用
Lactobacillus metabolites induces apoptosis of human tongue squamous carcinoma cells
【摘要】 目的:考察植物乳杆菌代谢产物对人舌鳞癌细胞系CAL-27细胞增殖的抑制作用和诱导细胞凋亡的作用。方法:植物乳杆菌23201分别经复原乳清培养基和复原乳清菊糖培养基培养,制备相应的代谢产物1(LPM1)和代谢产物2(LPM2);用MTT法检测细胞增殖抑制作用;通过吖啶橙染色法检测凋亡细胞的形态学变化;采用琼脂糖凝胶电泳法检测细胞DNA片段。结果:不同浓度LPM1和LPM2分别作用CAL-27细胞24 h,最大抑制率分别达到(44.2±3.3)%和(41±4.1)%;荧光显微镜下观察,凋亡细胞的细胞核不规则,凝聚呈新月形或马蹄形;琼脂糖凝胶电泳显示出典型的"梯"状DNA条带。结论:植物乳杆菌代谢产物可以抑制CAL-27细胞的增殖,呈剂量依赖性关系,并诱导CAL-27细胞凋亡。
【Abstract】 Objective: To investigate the effects of Lactobacillus plantarum metabolites on the proliferation and apoptosis of tongue squamous cell carcinoma CAL-27 cells.Methods: Lactobacillus plantarum metabolites 1and 2(LPM1 and LPM2) were obtained by culturing Lactobacillus plantarum 23201 in reconstituted whey medium and reconstituted whey-inulin medium respectively.MTT assay was used to test the inhibition effect of LPM1 and LPM2 on CAL-27 cells.Morphological changes of apoptotic cells were observed under fluorescence microscope after acridine orange(Ao) fluorescent staining.Agarose gel electrophoresis was used to detect the DNA fragments in CAL-27 cells after the treatment by LPM1 and LPM2.Results: After CAL-27 cells were treated with different concentrations of LPM1 and LPM2 for 24 h,maximum inhibition rates were(44.2±3.3) % and(41±4.1)% respectively.Condensed nuclei,horseshoe-like or crescent shapes of nuclei were observed in the LPM1 or LPM2 treated cells.The typical DNA ladders of the treated cells were detected by agarose gel electrophoresis.Conclusion: LPM may inhibit the proliferation and induce the apoptosis of CAL-27 cells in a dose-dependent manner.
