【摘要】 目的 研究MMP-3的基因多态性及其在胎膜组织上的表达情况与早产胎膜早破相关性。方法荧光定量PCR(SYBR Green法)检测MMP-3m RNA;Western blot法检测MMP-3蛋白表达量;MALDI-TOF质谱分析法对MMP3基因型检测分析。结果 早产胎膜早破组与足月胎膜早破组、正常足月组胎膜组织上MMP-3m RNA含量比较,差异有统计学意义;MMP-3蛋白表达量早产胎膜早破组、足月胎膜早破组和正常足月组分别为(0.76±0.10)、(0.40±0.06)和(0.25±0.03),早产胎膜早破组高于足月胎膜早破组及正常足月组,差异有统计学意义;早产胎膜早破组MMP-3(rs617819)C/C、G/C、G/G基因型分布频率、C等位基因分布频率与足月胎膜早破组相比,差异有统计学意义;MMP-3(rs35068180)A/A、A/-、-/-基因型分布频率、A等位基因分布频率早产胎膜早破组与其他各组比较,差异有统计学意义。结论 MMP-3基因多态性与早产胎膜早破发生存在一定相关性,MMP-3(rs35068180)A/A、A/-、-/各基因型、MMP-3(rs35068180)A等位基因有可能是早产胎膜早破的高危易感因素。更多还原

【Abstract】 Objective To examine genetic polymorphism of MMP-3 and its expression on the fetal membranes and their role in the pathogenesis of preterm premature rupture of membranes.Methods Fluorescence quantitative PCR was used to detect MMP-3 mRNA,and western blot was used to detect MMP-3 protein expression.MALDI-TOF mass spectrometry was used to detect MMP-3 single nucleotide polymorphisms.Results The premature rupture of membrane group had higher MMP-3 expression level(0.76±0.10)when compared with the full-term rupture of membrane group(0.40±0.06)and the full-term normal group(25±0.03).The distribution of MMP-3 rs617819 genotypes(C/C,G/C,G/G)and C allele was different between the preterm premature rupture of membrane group and the full-term rupture of membrane group.In addition,the distribution of MMP-3 rs35068180 genotypes(A/A,A/-,-/-)and A allele in the preterm premature rupture of membrane group was different from the distribution of the other two groups.Conclusion There was an association between with MMP-3 polymorphism and risk of preterm premature rupture of membranes.MMP-3 rs35068180 genotypes(A/A,A/-,-/-)and A allele may be genetic risk factors for preterm premature rupture of membranes.更多还原
