【摘要】 目的探讨米非司酮抑制紫杉醇诱导卵巢癌细胞株A2780、skov3耐药性的产生。方法紫杉醇诱导A2780、skov3过程中同时应用不同浓度的米非司酮,采用免疫组化法观察诱导后各细胞亚株中P糖蛋白(P-glyco-protein,P-gp)表达情况;检测各组细胞生长曲线;MTT法检测各细胞亚株的IC50;以罗丹明123(Rohdamin 123,Rh123)作为荧光探针应用流式细胞仪观察各细胞亚株胞内罗丹明含量;Western Blot法检测各细胞亚株中P-gp含量。结果诱导过程中应用米非司酮,使诱导后的A2780、skov3细胞株中P-gp表达水平下降。结论米非司酮可以抑制卵巢癌紫杉醇耐药性的产生。更多还原

【Abstract】 Objective To study the inhibitive effect of RU486 on resistance induced by taxol in ovarian cancer cells line A2780、skov3.Methods Immunohistochemical technique were used to observe the expression of P-glycoprotein(P-gp) in cells line A2780,skov3 treated with different concentration of RU486 and Taxol.Cells growth curve were detected.MTT method was used to study IC 50 in every cell lines.Rohdamin 123 was used as fluorescent probe to observe the effects of every cell lines.Western Blot was used to determine the expression of P-gp.Results The cell lines that treated by RU486 can decrease the expression of P-gp in cell lines.Conclusion RU486 can inhibit resistance of ovarian cancer cell lines induced by Taxol.更多还原
