2-脱氧-D-葡萄糖对人卵巢癌SKOV-3增殖的影响

【摘要】 目的探讨2-脱氧-D-葡萄糖(2-deoxy-D-glucose,2-DG)对人卵巢癌SKOV-3增殖及凋亡的影响。方法培养人卵巢癌SKOV-3细胞,将不同浓度2-DG(2.5 mmol/L、5 mmol/L、10 mmol/L、20 mmol/L、40 mmol/L)作用人卵巢癌SKOV-3细胞不同时间后,MTT检测各组细胞生长抑制率;Hoechst33258染色后,荧光显微镜下观察细胞形态变化;应用流式细胞仪检测细胞凋亡率及周期。结果不同浓度2-DG对SKOV-3细胞均有增殖抑制作用,与对照组相比差异有统计学意义(P<0.05),并且呈现剂量和时间依赖性;结果还显示2-DG可使细胞周期阻滞在G0/G1期;流式细胞仪凋亡检测结果显示2-DG可诱导人卵巢癌坏死或凋亡。结论体外试验中,2-DG对SKOV-3细胞增殖有抑制作用,且呈剂量和时间依赖性。

【Abstract】 Objective To investigate the influence of 2-deoxy-D-glucose(2-DG) on proliferation and apoptosis of human ovarian cancer SKOV-3 cells.Methods The human ovarian cancer SKOV-3 cells were treated with 2-DG in different concentrations(2.5 mmol/L 、5 mmol/L、10 mmol/L、20 mmol/L、40 mmol/L).Then MTT was used to detect the cell growth inhibition rate of each group at different time;Hoechst33258 staining assay was chosen to observe the morphological changes induced by 2-DG;the cell cycle and apoptosis were analyzed via flow cytometry.Results Different concentration of 2-DG can inhibit the proliferation of SKOV-3 cells in a dose and time-dependent manner,and there was significant difference compared with control group(P<0.05);Our results also showed 2-DG treated cells were arrested in G0/G1 phase;Apoptosis assay by Flow showed admonsted that 2-DG can induce cell apoptosis or necrosis.Conclusion 2-DG has inhibitory effect on the proliferation of SKOV-3 cells in vitro,and the effect is dose and time-dependent.
