【摘要】 目的:研究NDRG-1与上皮细胞钙粘蛋白(E-cadherin)在肝细胞癌组织中的表达和相关性,明确在肝细胞癌细胞中二者之间的调控关系。方法:实时定量PCR检测原发无转移肝细胞癌和原发伴转移肝细胞癌组织中NDRG-1和E-cadherin的表达。真核表达载体pcDNA3.3-NDRG1转染肝细胞癌HepG2细胞,Western blot检测NDRG-1和E-cadherin蛋白的表达。结果:与原发无转移肝细胞癌相比,原发伴转移肝细胞癌组织中的NDRG-1表达呈现显著上调,E-cadherin表达呈现显著下调,且二者呈显著负相关。转染后,NDRG-1蛋白的表达显著上调,而E-cadherin蛋白的表达显著下调。结论:NDRG-1和E-cadherin与肝细胞癌的转移相关,NDRG-1在肝细胞癌细胞中能够下调E-cadherin的表达。更多还原

【Abstract】 Objective: To research the expression and relationship of NDRG-1 and E- cadherin in hepatocellular carcinoma,and the regulation relationship between them in hepatocellular carcinoma cells. Methods: Real- time PCR was used to detect NDRG- 1 and E- cadherin mRNA in primary hepatocellular carcinoma without metastasis and primary hepatocellular carcinoma with metastasis. After the expression vector pcDNA3. 3- NDRG1 was transfected to HepG2 cell,the expression of NDRG- 1 and E- cadherin protein was deteced by Western blot. Results: The mRNA expression of NDRG- 1 in primary hepatocellular carcinoma with metastasis was much higher than that in primary hepatocellular carcinoma without metastasis,but the expression of E- cadherin was much lower,and there was a positive relationship between them. After transfection,the expression of NDRG- 1 protein increased significantly,and the expression of E- cadherin decreased significantly. Conclusion: The NDRG- 1 and E- cadherin gene have a relationship with metastasis of hepatocellular carcinoma,NDRG- 1 can down- regulate the expression of E- cadherin in hepatocellular carcinoma cells.更多还原
