褪黑激素受体Ⅰ型(MTR1)在乳腺癌组织中的表达及其临床意义

【摘要】 目的:探讨褪黑激素受体Ⅰ型(MTR1)在乳腺癌、正常乳腺组织中的表达及其临床意义。方法:收集140例乳腺组织样本,分为2组,乳腺癌组、正常乳腺组,每组70例,采用免疫组化染色(SP法)和WesternBlot方法检测2组MTR1的表达及其差异。结果:乳腺癌和正常乳腺组织的免疫组化染色反应强度综合评分分别为4.26±1.82,1.80±1.19(P=0.00),阳性表达率分别为81.4%、64.3%(χ2=5.201,P=0.023),MTR1在乳腺癌组织中的表达水平高于其在正常乳腺组织中的表达水平(P<0.05),差异有统计学意义。Western Blot检测结果为MTR1表达水平乳腺癌组(2.81±0.44)高于正常乳腺组(1.11±0.15),两组比较具有统计学意义(P=0.00),与免疫组化结果相一致。结论:MTR1在正常和病理的乳腺组织中均有表达,MTR1在乳腺癌组织中的高表达提示MTR1可作为乳腺癌诊断的分子标志之一和乳腺癌的潜在治疗靶点。

【Abstract】 Objective:To study the difference of expression level of MTR1 melatonin receptor in the tissue of human breast cancer and normal breast and its clinical significance.Methods: All 140 samples of breast tissue were divided into two groups each group with 70 cases: breast cancer and normal breast.Expression level of MTR1 was examined in two groups by Western Blot and immunohistochemical staining SP method.Results: Integrated assessment of positive results of breast cancer and normal breast were 4.26±1.82,1.80±1.19(P=0.00).The positive rate of expression of MTR1 was respectively 81.4% in the tissues of breast cancer and 64.3% in the tissues of normal breast(χ2=5.201,P=0.023<0.05).The expression level of MTR1 in the tissues of breast cancer(2.81±0.44) were higher than that of normal breast(1.11±0.15),(P=0.00).Conclusion: The MTR1 receptor was expressed both in the tissue of breast cancer and normal breast.The over expression of MTR1 receptor in the tissue of breast cancer which can be used as one of the molecular mark for diagnosis of breast cancer and potential target for breast cancer therapy.
