辐射对小鼠大脑微血管内皮细胞的作用
Effect of irradiation on cerebral microvascular endothelial cells
【摘要】 目的研究辐射单因素对小鼠大脑微血管内皮细胞的影响,并探索辐射对脑微血管内皮细胞的可能的调控机制。方法采用原代培养的小鼠大脑微血管内皮细胞,分为3组,辐射剂量分别为0,1,2 Gy。应用TUNEL染色观察小鼠大脑微血管内皮细胞形态学变化和凋亡情况。应用Western blot方法测定MAPKs家族ERK、JYN蛋白的磷酸化水平。结果随着辐射剂量增加细胞凋亡比例升高,以2 Gy辐射下的细胞凋亡最明显;ERK蛋白磷酸化水平在辐射剂量1 Gy时,出现下降趋势,随辐射剂量增大后呈现升高趋势,JNK蛋白磷酸化水平呈现升高趋势。结论辐射可引起小鼠大脑微血管内皮细胞凋亡,且随辐射剂量增加,细胞凋亡率随之上升。MAPK/ERK、MAPK/JNK信号转导通道可能参与辐射对小鼠大脑微血管内皮细胞凋亡的调节过程。
【Abstract】 Objective To investigate the effects of irradiation on cerebral microvascular endothelial cells and to explore its the potential mechanisms.Methods The primary culture of murine brain microvascular endothelial cells were divided into 3 groups and given 60Co at the dosage of 0 Gy,1 Gy,2 Gy,respectively.Cell morphology and the apoptotic cells of murine brain microvascular endothelial cells were observed by TUNEL method.And the expression of p-ERK and p-JNK was detected by Western blot.Results The apoptosis increased with the increase of radiation dose.The expression of p-ERK decreased at 1Gy radiation dose,and then increased.And the trend of expression of p-JNK was upwards along with the increasing radiation dose.Conclusion The radiation can lead to cell apoptosis,and there are positive correlations between the radiation dosage and the apoptotic rate.MAPK/ERK and MAPK/JNK signal transduction pathways may participate in the regulation of cell apoptosis under radiation conditions.
