西妥昔单抗抑制乳腺肿瘤细胞的增殖
Cetuximab suppresses proliferation of human breast cancer cells
【摘要】 目的研究西妥昔单抗对乳腺肿瘤细胞生长的影响。方法 MDA-MB-231细胞在2μg/mL西妥昔单抗刺激12h和24h后,收集细胞进行裂解。等量蛋白进行蛋白印迹法检测integrinβ5表达。软琼脂克隆形成实验检测西妥昔单抗对细胞生长的影响。结果西妥昔单抗明显降低integrinβ5蛋白表达。MDA-MB-231细胞在西妥昔单抗的作用下降低了克隆形成数量。结论西妥昔单抗能够下调integrinβ5表达,抑制乳腺癌细胞锚定非依赖生长,提示西妥昔单抗可能是重要的乳腺癌治疗靶向药物。
【Abstract】 Objective To examine the influence of cetuximab on the proliferation of breast cancer cells. Methods MDA-MB-231cells were exposed to vehicle alone as control or 2μg/mL cetuximab for 12hand 24h, and then cells were harvested and lysed.Lysates containing equal amounts of protein were Western blotting with integrinβ5antibody.Cells proliferation capacity was evaluated by using a colony formation assay.Results Cetux- imab dramatically decreased the levels of integrinβ5protein.Colony formation in soft agar by MDA-MB-231cells was effectively decreased by treatment with cetuximab.Conclusion We showed a novel mechanism of modulation of cell proliferation by cetuximab through down-regulation of integrinβ5in breast cancer cells.Cetuximab treat- ment prevents anchorage-independent colony formation in MDA-MB-231cells,which is critical for the design of targeted therapies against human breast cancer.
