凋亡因子capase-12在压疮早期损伤中的作用
Effect of apoptotic factor caspase-12 in early injury of pressure ulcers patients
【摘要】 [目的]分析凋亡因子caspase-12在压疮早期损伤中表达及动态变化,探讨压疮损伤发生的细胞分子机制,为临床预防压疮发生提供理论依据。[方法]将40只雄性SD大鼠随机分为5组(8只/组),对照组大鼠不施加压力,实验组模拟临床压疮发生条件,施压2h,放松0.5h,为1个周期,每天进行3个周期,根据施压周期的不同又分为1c组、3c组、6c组、9c组。运用HE染色观察组织的病理学变化,免疫组织化学法检测受压肌肉组织中内质网特异性凋亡因子caspase-12的动态变化。[结果]光镜下观察到,实验组随着周期延长,受压肌肉组织呈现逐渐退化的变化;免疫组织化学结果显示,实验组凋亡因子caspase-12蛋白表达均高于对照组,差异有统计学意义(P<0.05),阳性反应主要发生在胞浆和细胞核表达。[结论]压疮早期损伤激活了内质网应激反应,且激活凋亡因子caspase-12诱导细胞发生损伤。
【Abstract】 Objective:To analyze the expressions and dynamic change of apoptotic factor caspase-12 in early injury of pressure ulcers patients,and probe into the molecular mechanism of pressure ulcers,so as to provide theory evidences for prevention of clinical pressure ulcer.Methods:A total of 40 male SD rats were divided into 5 groups randomly,eight rats in each group,control group rats without applying pressure,the experimental group rats simulated clinical pressure ulcer conditions,exerting pressure for 2 h,relax for 0.5 h,3 cycles for every day.According to different pressure cycles the experimental group was divided into 1c,3c,6c,9c group.By using HE staining to observe tissues pathological changes and immunohistochemical method to detect the dynamic changes of endoplasmic reticulum-specific apoptotic factors caspase-12 in pressured muscle tissue.Results:Compared with the control group,the results under the light microscope of HE staining showed,the compression muscle tissue of the experiment group showed a gradual degradation in the pathological changes with the extended cycle.The immunohistochemical results indicated that the protein expression of caspase-12 was elevated in the experiment group compared with the control group(P<0.05).Conclusion:Early injury of pressure ulcers activated endoplasmic reticulum stress response and activate the apoptotic factors caspase-12 and then to induce cell damage.
