可口革囊星虫不同组织同工酶的比较
The comparison of isozymes in different tissues of Phascolosoma esculenta
【摘要】 采用聚丙烯酰胺凝胶电泳(PAGE)技术,研究了可口革囊星虫5种组织(收吻肌、吻、肠、肾管及体壁)10种同工酶(ADH、GCDH、SDH、EST、LDH、ME、POD、SOD、MDH、CAT)的表达。结果表明:除CAT外,其余9种同工酶均得到了较为清晰的酶谱,除SOD和MDH在5种组织中表型相同外,其它7种同工酶的表达均具有明显的组织特异性。SDH-2、EST-1、LDH-1和POD-1为收吻肌所特有;ADH-3、ADH-4、GCDH-2、GCDH-4、GCDH-5、EST-10和LDH-6均为吻所特有;EST-4为肾管所特有。在肠中,ADH和GCDH均没有检测到活性。初步探讨了5种组织中各种同工酶的分布与活性及其与各组织生理功能的关系。
【Abstract】 Polyarylamide gel electrophoresis was used to detect the expression of isozymes in 5 tissues of Phascolosoma esculenta such as introvert retractor muscle,introvert,intestine,nephridium and somatocyst.Isozymers like ADH,GCDH,SDH,EST,LDH,ME,POD,SOD,MDH and CAT were analyzed and the results were showed as follows: except CAT isozyme the rest isozymes present specified clear and steady bands in different tissues,and seven of them(ADH,GCDH,SDH,EST,LDH,ME and POD) exhibited apparent tissue specificity,except SOD and MDH.In the five tissues,enzymes like SDH-2,EST-1,LDH-1 and POD-1were expressed in the introvert retractor muscle,enzymes like ADH-3,ADH-4,GCDH-2,GCDH-4,GCDH-5,EST-10 and LDH-6 were expressed in the introvert,and EST-4 was expressed in the nephridium,all the isozyme bands above were experssed one and only.The activity of ADH and GCDH had not been detected in the intestine of Phascolosoma esculenta.In the paper,the distributing and activity of ten isozymes in 5 tissues and the functions of those tissues were discussed.
