SAHA对TGF-β诱导人胚肺成纤维细胞α-SMA蛋白及前胶原蛋白mRNA表达的影响
Effect of SAHA on TGF-β-induced α-SMA and Procollagen Protein Expression in Human Embryonic Lung Fibroblast Cells
【摘要】 目的:探讨在转化生长因子-β1(TGF-β1)刺激下,组蛋白去乙酰化酶抑制剂辛二酰苯胺异羟肟酸(SAHA)对人胚肺成纤维细胞系HELF向肌成纤维细胞(MF)分化时,α-SMA蛋白及前胶原蛋白mRNA表达的影响。方法:体外培养人胚肺成纤维细胞系HELF,并根据不同的实验目的分为空白对照组,TGF-β1处理组以及SAHA干预组。细胞处理结束后,用Western blot检测α-SMA表达,RT-PCR检测I、III型前胶原的mRNA表达水平。结果:空白对照组中几乎无α-SMA蛋白表达,TGF-β1处理后α-SMA水平显著增高,而SAHA能有效降低α-SMA的水平。SAHA孵育24h后,能够明显抑制TGF-β1刺激后的细胞表达I型和III型前胶原蛋白mRNA,并且具有明显的剂量依赖性。结论:SAHA能降低TGF-β1诱导HELF细胞向MF转化时α-SMA表达以及前胶原蛋白表达。
【Abstract】 Objective: To investigate the effect of suberoylanilide hydroxamic acid(SAHA) on transforming growth factor-β1(TGF-β1)-inducedα-SMA and procollagen protein expression in human embryonic lung fibroblast cells(HELF).Methods: HELF cells were cultured in vitro and divided into Control group,TGF-β group and SAHA groups according to the different purpose.After treated,Western-blot was used to assessα-SMA expression,and the mRNA levels of I,III procollagen in HELF cells were detected by RT-PCR.Results: α-SMA expression almost could not be detected in the control group,while after TGF-β1 treatment.The level of α-SMA was significantly,and SAHA could also reduceα-SMA expression.After 24 h incubation of SAHA,mRNA expression of type I and type III procollagen can be inhibited by stimulation of TGF-β1 in dose-dependent manner.Conclusion: SAHA can inhibit TGF-β1-induced α-SMA and procollagen expression.
