RASSF1A对食管癌细胞增殖影响的研究

【摘要】 目的:观察外源性RASSF1A基因对食管癌细胞的增殖作用并探讨机制。方法:将包含RASSF1A基因的质粒转染食管癌细胞EC9706,建立稳定转染细胞克隆,Western blot检测RASSF1A基因表达,通过细胞生长曲线检测细胞生长活性和增殖能力、流式细胞仪检测对细胞周期的影响、裸鼠移植瘤实验检测转染细胞的体内成瘤特性。结果:稳定表达RASSF1A基因的EC9706细胞中RASSF1A蛋白表达增加;细胞生长速度明显减慢;细胞周期中G1/G0期比例明显增加,S期比例减少;EC9706细胞的裸鼠致瘤能力被抑制。结论:RASSF1A基因能显著抑制食管癌细胞在体内外的生长,其机制可能与该基因诱导凋亡、抑制增殖作用有关。

【Abstract】 Objective: To investigate the suppressive roles on tumorigenesis effects of RASSF1A in human esophageal carcinoma cells in vivo and in vitro and the molecular mechanisms.Methods: Vector pcDNA3.1(+) and pcDNA3.1(+)-RASSF1A were transfected into EC9706 cells by lipofectamine 2000.The expression of RASSF1A protein was examined by Western Blot.The cytobiologic character of the RASSF1A transfected EC9706 cells were probed by cell growth curve,cell cycle analysis and xenograft of nude mice.Results: Western Blot showed that RASSF1A had high expression,and cell viability and growing speed were decreased obviously in RASSF1A transfected EC9706 cells.The progression of the cell cycle was arrested in G1 phase remarkably,and the tumor formation in mude mice was inhibited.Conclusion: The results indicated that RASSF1A may function as a tumor suppressor gene to play an important suppressed role in origination,progression and metastasis of esphageal carcinoma.
