冷藏真空包装脆肉鲩鱼特定腐败菌的分析
Specific spoilage organism of vacuum-packed crisped grass carp during chilled storage
【摘要】 利用16S rDNA克隆文库及限制性内切酶片段长度多态性(Restriction Fragment Length Polymorphism,RFLP)技术,探讨了导致冷藏真空包装脆肉鲩鱼的特定腐败菌,为研发保鲜加工技术及质量监控提供参考及依据。通过提取冷藏腐败的真空包装脆肉鲩鱼肉的细菌总DNA,采用细菌16S rDNA通用引物27F/1492R进行PCR扩增,构建16S rDNA克隆文库,经RFLP分析、测序及序列比对后构建系统发育树。RFLP分析结果表明,冷藏腐败的真空包装脆肉鲩鱼肉中有12种不同分型的细菌种类,其中分型1和2占克隆子数的58.92%,是该条件下的优势菌;序列比对分析及构建的系统发育树显示,包括分型1和2的大部分克隆子的序列与气单胞菌属(Aeromonas)细菌的16S rDNA序列有最大的相似性,表明真空包装脆肉鲩鱼冷藏的特定腐败菌是气单胞菌(Aeromonas sp.)。
【Abstract】 Specific spoilage organism of vacuum-packed crisped grass carp during chilled storage were analyzed by 16S rDNA clone library and restriction fragment length polymorphism(RFLP) to provide evidence for processing,preservation and quality control. The total bacterial DNA was extracted from spoilage sample,then 16S rDNA genes were amplified by PCR using universal primers of 27F/1492R to construct 16S rDNA clone library. The library was analyzed by RFLP,sequencing,blast and then constructed phylogenetic tree. RFLP results indicated there were 12 types of bacteria which type 1and 2 were predominant(58.93% of total clones) in spoilage fish. 16S rDNA sequences analysis and phylogenetic tree results showed most clone seuqences include clones of type 1 and 2 were similar with16S rDNA seuqences of Aeromonas. These results demonstrated Aeromonas sp. was specific spoilage organism of vacuum-packaged crisped grass carp during chilled storage.
