氯霉素ELISA检测试剂盒评价技术

【摘要】 酶联免疫吸附实验(ELISA)试剂盒作为多种实验要素的集合,已经广泛应用于日常检测和实验研究中。为了解国产氯霉素试剂盒的有效性和可靠性,选择一种市售试剂盒,从灵敏度、特异性、准确度、重复性、检出限、耐变性、批间变异等多个角度对其进行了技术评价。该试剂盒在测试范围内线性回归良好,相关系数R2可达0.9945,与结构相似的棕榈氯霉素、甲砜霉素、氟甲砜霉素的交叉反应率均小于0.1%,批间变异系数小于20%,鸡肉、虾肉的平均回收率为100.9%、100.7%,相应的检测限是0.077、0.128ng/mL,可以满足"2012年度动物及动物产品兽药残留监控计划"的限量标准(0.3ng/mL)要求。

【Abstract】 The enzyme linked immunosorbent assay is a commonly used method of immobilized enzyme immunoassay.As a set of experimental elements,it has been widely used in daily detection and experimental study,in order to understand the effectiveness and reliability of domestic Chloramphenicol kit,the authors chose a commercially available kit and evaluated the performance of ELISA in terms of sensitivity,specificity,accuracy,reproducibility,the detection limit,stability and inter-assay coefficients of variation.The results showed that the ELISA had a regression line of y=-17.001Ln(x)+35.241 and correlation coefficient 0.9945.It showed less than 0.1% cross-reactivity with Chloramphenicol palmitate thiamphenicol and florfenicol,which was similar to chloramphenicol in structure.The inter-assay coefficients of variation is less than 20%,and the rate of recovery for CAP from chicken muscle and shrimp flesh was 100.9%,100.7%,the corresponding the detection limit was 0.077,0.128 ng/mL,it conformed the limit(0.3 ng/mL) which referred to the "monitoring program of veterinary drug residues in animal and animal products of 2012" and can be used as commercial kit.
