【摘要】 对微生物学实验"产生蛋白酶菌株的筛选、鉴定、培养和选育"进行了教学改革,改变了以往的内容安排以及考核方式,并将该实验综合为一个统一的实验模块.模块是由8个紧密相连的实验所组成的一个综合性实验模块,其每个实验的具体内容分别为:玻璃器皿的清洗、包扎,培养基的配制及灭菌,产蛋白酶菌株的筛选、纯化及保藏,产蛋白酶菌的革兰氏染色及镜检,产蛋白酶菌的PCR鉴定,16S rDNA序列的比对及其分析,产蛋白酶细菌的生理生化鉴定,产蛋白酶菌的培养优化实验以及其诱变选育.更多还原

【Abstract】 Contents and examination forms of teaching comprehensive experiment of microbiology,which focuses on isolation,identification,cultivation and breeding of protease-producing strains,are introduced in this paper.The comprehensive experiment was composed of eight closely linked parts,which were as follows: clean and pack of glassware,and preparation and sterilization of media;screening,purification and preservation of protease-producing strains;gram stain and examination of microscope;PCR identification of strains;alignment and analysis of 16S rDNA sequence of the strains;identification of physiological and biochemical characters of the strains;optimization of the culture medium and mutation breeding of the strains.更多还原
