交叉引物等温扩增技术检测志贺氏菌

【摘要】 建立一种新型志贺氏菌简洁快速检测方法——交叉引物恒温扩增检测方法。针对志贺氏菌的四个血清型设计保守的特异性引物及探针,建立交叉引物等温扩增法并应用免疫金标试纸条进行检测。用6株志贺氏菌,32株近源菌进行特异性试验;通过纯菌液计数、干扰菌试验检测进行灵敏度验证。建立方法具有较好的特异性,增菌液及干扰菌检测灵敏度均为103cfu/mL。建立的交叉引物恒温检测方法灵敏度高,可满足对志贺氏菌进行现场快速筛查目的。

【Abstract】 To establish a new rapid method for detection of Shigella———cross-primer amplification.On the basis of sequence analysis of the Shigella,specific primers and probe were designed,we developed cross-primer isothermal amplification method.The specificity of the method was evaluated by 6 strains of Shigella and 32 negative strains of Enterobacteriaceae.The sensitivity of the method was evaluated by testing thepure bacteria counted and interference bacteria test.The method established in this article is highly specific and sensitive.The limit of decetion is 102 cfu/mL for pure bacterial culture and interference bacteria test.The method is highly sensitive,easy to operate and useful for rapid screen of Shigella in the port or underdeveloped areas.
