大豆提取液中大豆黄素的紫外分光光度测定法
UV Spectrometry Method to Determine Daidzein in Soybean Extract
【摘要】 紫外分光光度法(248 nm)测定大豆提取液中的大豆黄素浓度与HPLC法测得的值有显著差异(t检验法)。以HPLC法测得的值为可信值,用函数的方法对紫外法标准曲线进行矫正,矫正后的紫外法标准曲线方程为y=0.706 1x+0.012 5,回收率为99.128%,精密度为5.387%,用此方程测得的样液中大豆黄素浓度与HPLC法测得的值无显著差异(t检验法)。
【Abstract】 There was significant difference between the two daidzein values in the soybean extract determined by UV(at 248 nm)and HPLC(t test,>t0.01).UV standard curve was corrected by mathematic method and HPLC values as truth-values,a new relationship between concentration(y)and absorbency(x),y=0.706 1x+0.012 5,was established,with no significant difference between UV and HPLC,the recovery of 99.128 % and the relative standard deviation(RSD)of 5.387 % for daidzein detection.
