桔梗皂苷D对氧化型低密度脂蛋白诱导的内皮细胞氧化损伤的作用
Effect of Platycodin D on OxLDL-induced Oxidative Injury of Endothelial Cells
【摘要】 为检测桔梗皂苷D对氧化型低密度脂蛋白(OxLDL)诱导的内皮细胞氧化损伤的作用,利用OxLDL诱导人脐静脉内皮细胞(HUVECs),建立细胞氧化损伤模型,分别以0.5、0.75、1mg/mL桔梗皂苷D(PD)处理,测定培养液一氧化氮(NO)和丙二醛(MDA)水平;实时荧光定量聚合酶链式反应(qRT-PCR)检测细胞动脉细胞黏附分子(VCAM-1)和细胞黏附分子(ICAM-1)的表达;同时进行单核细胞与内皮细胞的黏附实验。结果表明:PD能显著增大细胞培养液NO浓度(P<0.01),显著降低MDA水平(P<0.01),减少VCAM-1和ICAM-1分子的表达(P<0.01)及单核细胞与内皮细胞的黏附作用(P<0.01)。说明PD能抑制OxLDL诱导的内皮细胞氧化损伤。
【Abstract】 The effect of platycodin D(PD) on human umbilical vein endothelial cells(HUVECs) were evaluated in vitro.OxLDL at 50 mg/L was used to induce oxidative injury in HUVECs to establish a cell model of oxidative injury.The cells were pre-treated with various concentrations of PD(0.5,0.75 mg/mL and 1 mg/mL).The levels of nitric oxide(NO) and malonaldehyde(MAD) in the culture medium were determined.The mRNA expression of vascular cell adhesion molecule-1(VCAM-1) and intercellular cell adhesion molecule-1(ICAM-1) in endothelium cells were also analyzed as well as the adhesion of monocytes to endothelial cells.The results showed that PD treatment significantly increased NO levels(P < 0.01) but decreased(P < 0.01) MDA levels in the medium of endothelial cells.Moreover,PD significantly downregulated mRNA expression levels of VCAM-1 and ICAM-1 in endothelial cells(P < 0.01) but also inhibited the monocyte adhesion to endothelial cells(P < 0.01).These findings suggest that PD can inhibit OxLDL-induced oxidative injury in endothelial cells.
