矢车菊素-3-葡萄糖苷抑制KKAy糖尿病小鼠PTEN表达
Cyanidin 3-Glucoside Ameliorates Insulin Sensitivity through Down-regulation of PTEN in Diabetic KKAy Mice
【摘要】 通过在KKAy小鼠膳食中添加花色苷Cy-3-g(1g/kg膳食)干预12周,分析Cy-3-g对小鼠血糖、血清胰岛素含量的作用,并进一步检测肝脏、肌肉、内脏脂肪组织PTEN mRNA、蛋白以及磷酸化Akt蛋白的表达。结果表明:Cy-3-g膳食干预可以改善KKAy小鼠胰岛素敏感性,抑制肝脏、肌肉、内脏脂肪组织PTEN表达而增强Akt磷酸化,提示富含Cy-3-g的花色苷提取物可能是通过抑制PTEN表达而激活PI3K/Akt通路,从而改善小鼠胰岛素抵抗。
【Abstract】 Cyanidin-3-O-β-glucoside(Cy-3-g)-rich foods have been reported to ameliorate the onset of insulin resistance,but the underlying mechanism has remained unclear.PTEN(phosphatase and tensin homologue deleted on chromosome 10) plays a negative role in insulin signaling-PI3K(phosphatidylinositol 3-kinase)/Akt(protein kinase B),and its inhibition improves insulin sensitivity.In order to gain insight into the effect of Cy-3-g anthocyanin on insulin sensitivity and the role of PTEN,KKAy mice were fed AIN-93G diet supplemented with or without Cy-3-g(1 g/kg diet) for 12 weeks.Our results demonstrated that Cy-3-g ameliorated insulin sensitivity,and inhibited the PTEN expression in liver,muscle and viscera adipose tissues following the activation of Akt(Ser473) in mice.These findings provided evidence for the role of PTEN in PI3K/Akt(Ser473) pathway,thus suggesting that the possible mechanism may be the contribution of Cy-3-g to insulin signaling.
