乌贼墨肽聚糖对前列腺癌PC-3、DU-145细胞作用机制研究
Apoptosis Induction and Underlying Mechanism of Peptidoglycan from Cuttlefish Ink on Prostate Cancer PC-3 and DU-145 Cells
【摘要】 目的:研究乌贼墨肽聚糖对前列腺癌PC-3和DU-145细胞凋亡的影响,以及细胞凋亡的机制。方法:用HE染色观察肽聚糖作用后细胞形态变化,流式细胞术检测细胞早期凋亡率,免疫组化检测肽聚糖作用前后细胞中COX-2和VEGF蛋白阳性表达情况,原位杂交检测肽聚糖作用前后细胞中Bcl-2 mRNA和Caspase-3 mRNA表达的变化。免疫印迹检测肽聚糖作用前后细胞中COX-2、VEGF、Bcl-2和Caspase-3的蛋白表达情况。结果:肽聚糖作用后的细胞出现凋亡的形态学改变,流式细胞术检测后发现,早期凋亡细胞随着作用时间和药物质量浓度的增加而增加,两种细胞中的COX-2和VEGF蛋白表达量下降,Bcl-2 mRNA的表达量减少,而Caspase-3 mRNA表达量增加。免疫印迹结果显示,COX-2、VEGF和Bcl-2蛋白表达量下降,Caspase-3蛋白表达量增加。结论:乌贼墨肽聚糖可以诱导前列腺癌PC-3和DU-145细胞的凋亡,机制可能是下调COX-2、VEGF蛋白和Bcl-2 mRNA的表达;上调Caspase-3 mRNA的表达。
【Abstract】 Objective: To explore the antitumor effect of cuttlefish ink peptidoglycan(PGN) on PC-3 and DU-145 cells and its mechanism in vitro.Methods: After PC-3 and DU-145 cells were treated by cuttlefish ink PGN,HE staining was used to observe cell growth condition and morphology,FCM was used to observe cell apoptosis,immunocytochemical staining(SABC) was used to analyze the expression of COX-2 and VEGF proteins and in situ hybridization was used to analyze the mRNA expression of Bcl-2 Caspase-3.The expression of COX-2,VEGF,Bcl-2 and Caspase-3 in the two human prostate cancer cell lines were also examined using western blot.Results: Morphological features of apoptosis,and the early stage of apoptosis in treated cells was observed in a dose-dependent manner.Moreover,cuttlefish ink peptidoglycan exposure for 24 h could result in a decease in the expression of COX-2,VEGF and Bcl-2 and an increase in the expression of Caspase-3.Conclusion: Cuttlefish ink peptidoglycan can induce the apoptosis of PC-3 and DU-145 cells possibly by down-regulating the expression of COX-2 and VEGF proteins and Bcl-2 mRNA and up-regulating the expression of Caspase-3 mRNA.
