浙贝母总生物碱提取分离工艺研究

【摘要】 目的以提取物中浙贝母总生物碱含量达到50%以上为目标,优选浙贝母总生物碱的提取分离工艺。方法采用酸水提、醇提及大孔树脂初步富集等方法提取分离浙贝母总生物碱,确定最优提取分离工艺。结果浙贝母总生物碱的提取分离工艺为6倍量2%盐酸溶液提取2次,每次1h,10%NaOH调pH值至9.0,用等体积二氯甲烷萃取4次。优选的提取分离工艺可使浙贝母总生物碱含量达到50%以上。结论该工艺能明显提高浙贝母总生物碱的纯度,且稳定可行,可为浙贝母总生物碱工业化大生产及新药研发奠定基础。

【Abstract】 Objective To optimize the extraction and separation process of the total alkaloid from thunberg fritillary bulb by targeting over 50% of the total alkaloid from thunberg fritillary bulb.Methods The acid water extraction,alcohol extraction and macroporous resin concentration methods were adopted to extract and separate the total alkaloid from thunberg fritillary bulb and determine the optimal extraction and separation process.Results The extraction and separation process of the total alkaloid from thunberg fritillary bulb:extraction twice with 2% hydrochloric acid solution of 6 times amount as the herbal material,1 h each time,pH 9.0 adjusted by 10% NaOH,extraction for 4 times with methylene chloride of equal volume.The optimized extraction and separation process could reach 50% of the total alkaloid from thunberg fritillary bulb.Conclusion This process apparently improves the purity of the total alkaloid from thunberg fritillary bulb and is stable and feasible.Hence this process lays the foundation of the industrialization production of the total alkaloid from thunberg fritillary bulb and the research and development of new drug.
